Annotation
Enzymes of GH families 16, 28 and 72 belong to hydrolases/transglycosylases which are essential during reorganization of cell walls. Depending on research stadium, work will be focused on bioinformatic analysis of nucleotide and protein databases, preparation of transformed Pichia pastoris strains carrying synthetic genes, expression of enzymes and their purification and characterization with primary focus on their substrate specificities, structural studies, expression and function of individual isoforms in cell walls of plants and yeasts. Such a focus of work requires the use of a combination of methods of bioinformatics, computational chemistry, molecular biology, enzymology and several sophisticated analytical methods.
