Complex approach to the investigation of cell migration and invasion in 2D and 3D environment with quantitative output

Annotation:Metastasis of primary tumors connected with migration and invasion of cancer cells into surrounding tissue drastically reduces chances of survival of cancer patients. Therefore, molecules involved in these processes present potential candidates for anti-cancer therapy targets. The aim of the project is to use the powerful microscopy setup at the Institute of Virology for the development of optimized 2D and 3D cell migration and invasion assays to study these processes in the controlled environment. At the same time, we want to establish a reliable method for processing and quantification of 2D and 3D assay results, allowing their comparison and assessment of the relevance of 2D assay results for cell migration and invasion in more physiological conditions of 3D environment. Tumor hypoxia marker CA IX will be used to test the efficiency and reliability our approach.

Study of molecular mechanisms of carbonic anhydrase IX regulation and significance of its expression in colorectal cancer.

Annotation:Colorectal cancer (CRC) is one of the most common malignancies worldwide characterized by metastatic dissemination of primary tumor and high mortality of patients. Carbonic anhydrase IX (CA IX) is a well known marker of hypoxia, and its expression significantly correlates with poor prognosis and resistance to chemotherapy. Interestingly, the expression of CA IX in CRC only partially overlaps with the hypoxia-induced signaling pathway driven by transcription factor HIF-1. Thus, activation of additional factors and/or signaling pathways (c-Met/HGF, Cox-2/PGE2, β-catenin/APC) affecting within tumor microenvironment is feasible. The main goal of this project is to elucidate mechanisms regulating the expression of CA IX in CRC. Moreover, we intend to evaluate clinical significance of CA IX as a CRC marker with emphasis on its correlation with markers of progression (Cox-2) and metastasis (c-Met). We suppose that such a combination of biomarkers will increase their individual clinical significance.
