Improvement of therapeutic interventions in prevention/treatment of diabetic complications and chronic inflammatory disorders represents an urgent challenge. This work comprises a multidisciplinary approach based on the knowledge of relevant molecular factors, namely the role of the polyol pathway and oxidative stress in the etiology of the mentioned pathologies, combined with medicinal chemistry approaches in drug design, including in silico modeling and virtual screening. The experimental part will focus on the development of novel pharmacologically applicable aldose reductase inhibitors endowed with high efficacy and selectivity, good bioavailability and negligible toxicity is envisaged. Moreover, the results may contribute to understanding molecular mechanisms of glucose toxicity in the etiology of diabetic complications and oxidative stress related mechanisms in chronic inflammation.
