Immobilized and co-immobilized whole-cell biocatalysts containing enzyme cascades for the production of chemical specialties

The aim of the dissertation is to acquire new knowledge within the research of innovated immobilized and co-immobilized whole-cell biocatalysts based on viable recombinant microorganisms for production of precursors for synthesis of bioactive substances as well as development of methods for their characterization. A cascade of redox enzymes coexpressed in bacteria will be utilized as well as recombinant cells with overproduced individual cascade enzymes including enzymes from the Baeyer-Villiger monooxygenase family and the cells will be stabilized by immobilization and co-immobilization in polyelectrolyte particles and capsules. Advanced techniques for characterization of changes in cell physiology, micro- and nanostructure of immobilizing materials and optimization of the biocatalytic efficacy of immobilisates will be used in collaboration with leading domestic and foreign institutes.
